This is a Senior Scientist level position within the Reagent Generation team in the Biologics Discovery group of Janssen BioTherapeutics (JBIO). The Biologics Discovery group is responsible for discovering, characterizing and analyzing lead molecules for biotherapeutic drug development across all therapeutic areas of J&J. This position supports JBIO through the design and production of protein, cell, DNA, other reagents to enable discovery immunization campaigns and the subsequent screening of therapeutic panels. The successful candidate will possess expertise in recombinant protein production and mammalian cell engineering. The position also requires data analysis, interpretation of the results, preparation of reports and effective communication of results to management and program teams. This position may also include responsibilities for the supervision and development of associate scientists. As a Senior Scientist you will:
- You will collaborate with project teams from all therapeutic areas to identify the most promising and appropriate designs for immunogens and screening reagents.
- Produce and oversee the production of protein, cell, and DNA reagents to support antibody generation and screening for biotherapeutic drug discovery.
- Provide oversight and mentorship to junior group members and external organizations.
- Characterize the quality of protein, cell, and DNA reagents, review biophysical data from external organizations, and collaborate with other functional groups within JBIO to ensure immunogens are representative of native targets.
- A minimum of a PhD is required
A minimum of 2 years of post-doctoral OR at least 2 years of industry experience is required
Expertise in the expression, purification, and characterization of proteins from numerous hosts including mammalian cells (HEK, CHO, NS0) is preferred
Expertise in mammalian cell engineering including the production of monoclonal and stable pools over-expressing a specific target of interest, as well as production of gene-edited cell lines by CRISPR-Cas9 is required
Expertise in flow cytometry to analyze cell surface as well as intracellular target expression is required
Proficient use of public databases to extract relevant data – sequences, structural domains, signal peptide analysis and design is required
Protein engineering experience as well as understanding of structure of antibodies and antibody-like molecules including single domain/VHH, scFv, Fab, etc is preferred
Advanced understanding of structural biology particularly as it pertains to the design of protein sub-domains, fusion proteins, and chimeras is preferred
Experience generating engineered lines from B cell lymphoma lineage is preferred
- Experience in fluorescence activated cell sorting (FACS) preferred
Working understanding of immunology, physiology of immune response in different pre-clinical models preferred
