Janssen Research & Development, a division of Janssen Pharmaceutica NV, has opened up an excellent Computational Scientist position at our site in Beerse, Belgium. We are seeking a highly motivated and passionate Computational Scientist (Biomedical engineer; IT programmer with a strong background/interest in physiology) (m/f) to join our multinational Global Safety Pharmacology research group.


Safety pharmacology is a discipline within drug discovery/development that helps to select compounds based on their safety profiles. The field makes use of different in vitro, ex-vivo and in vivo models to determine potential drug-side effects. Currently there is a high interest to develop computer based (in silico) models for the prediction of side effects on the heart. The input for these in silico models is primarily based on in vitro experiments, and the output should give information on the potential of drugs to induce rhythm disturbances of the heart. The governmental regulators (in particular the US-FDA) have proposed the use of in silico models for so called cardiac “action potentials” based on the published O’Hara Rudy model (O’Hara at al.; 2011, PloS Comput. Biol. 7:e1002061. Doi: 10.1371/journal.pcbi.1002061). The current regulatory proarrhythmia safety testing paradigm (S7B) will be replaced shortly by a new more comprehensive paradigm. The CiPA (Comprehensive in vitro Proarrhythmia Assay) proposes to evaluate drug effects on multiple cardiac ion channels in vitro, use these data in a predictive in silico model of the adult human ventricular myocyte and confirm in vitro ion channel and in silico data in a human stem-cell derived cardiomyocyte assay (http://cipaproject.org/about-cipa/ ).  Available open source codes have already been made available by e.g. the FDA (including R and C codes; https://github.com/FDA/CiPA ).

 

Key job responsibilities:

  • Development of mechanism-based modelling and simulation and assist in building a practical in silico strategy for Safety Pharmacology
  • Building script/web-based application for a user-friendly tool to perform in silico simulation of cardiac ventricular cardiomyocytes for predicting risk of TdP of discovery compounds
  • Building tools to help chemists for SAR strategy of new compounds
  • Pursue, develop and establish fit for purpose protocols & techniques to support de-risking and development research based on new ideas and by building on existing computational models
  • Enable time-appropriate decision making for early stages of biological programs and for late stage compounds in development
  • Expected to draft, execute and serve as lead author on research articles, book chapters, reviews and internal reports
  • Involvement in translational research: correlation, prediction clinical observations from existing pre-clinical data
  • Expected to attend in house seminars and local science meetings within the field where necessary



Qualifications

Education & experience

·         PhD/Dphil in Computer Science, Biomedical Engineering, Physics or related discipline, together with relevant experience

·         The applicant can work independently but is also a team player. The applicant has an affinity to cell-physiology (Biology)

·         Experience/knowledge or strong interest in cardiac modelling and simulation

·         The applicant has a background in web- based applications

·         The applicant should be familiar with R, C codes, Matlab, etc..


Competencies

·         Preferably have strong background in models in cardiac simulations, high performance computing, machine learning and cardiac physiology

·         Excellent communication, reporting, planning, presentation (internal & external) and team interaction skills, self-motivation, proactivity and the ability to work both independently and as part of a multi-disciplinary and multi-national team

·         Ability to build bridges between colleagues, disciplines and departments

·         Advanced hands-on programming and scripting skills

Candidates must have a reasonable understanding of English (both written and spoken)
