The High Throughput Experimentation (THE) Director will lead the formation and execution of a high throughput experimentation team serving small molecule API customers in discovery and development. The incumbent will have responsibility for establishing and leading a team of scientists to provide expert support in “de-bottlenecking” difficult chemical transformations as well as enabling previously inaccessible chemical space in a robust and scalable manner to ensure processes amenable for scale-up are developed as soon as possible for successful candidates.
The incumbent anticipates future directions and challenges in the field of Organic Chemistry with an appreciation for deep chemical understanding and the challenges of process scalability foresight to ensure the HTE capabilities are maintained/expanded to support future directions.
Approximate Percentage of Time
Establish HTE team and capabilities scope:
  • The leader, in collaboration with leadership in SM discovery and development, will define the scope of capabilities and support the team will look to provide for technology and screening deployment. The scope should include clear endpoints for the services relevant to the requesting groups.
  • The leader will establish a framework for prioritization of effort across discovery and development requests.
  • The leader will be tasked with establishing areas of screening expertise in catalysis, biocatalysis, and reaction optimization.
  • Screening equipment will be identified and prioritized for purchase.
  • After the initial establishment of the HTE team and capability these responsibilities will become part of operational support.  The HTE leader will continuously define and explore new state-of-the-art areas of chemistry and build business cases to incorporate them into the HTE capability on an ongoing basis.

Operational Support:

·         The leader will be responsible for identifying the staff/talent required to execute on the screening areas of expertise and on-board them appropriately (internal/external).

·         The leader will be responsible for setting the vision and direction for the workflows – how the HTE group will receive requests, perform the workflows and provide the output back to the relevant requesting groups. The lead will also provide direction and strategy on appropriate level of screening support and scalability with the relevant requestors’ level of need in mind – workflows for providing access to novel chemical space will be different than workflows requiring catalyst optimization for scale-up as will the screening equipment required.

·         The lead will establish mechanisms to virtually increase the capacity of the HTE group via external partnering.

·         The lead will establish a data management and utilization framework to most efficiently capture and evaluate data in an automated/ML manner to enable faster and better screens to minimize the successive number of screening rounds needed. 


Project Execution Support:

·         The leader will establish a program for developing/implementing standard panels for screening of common transformations (i.e. Suzuki, KRED reductions, solvent screen) to enable fast turnaround for initial screening rounds serving the discovery space.

·         For more difficult transformations or those requiring a deeper level of chemical understanding additional manually designed screens can and will be performed.  The leader will deliver a framework for this level of project support and ensuring proper expertise is accessed within the HTE team to ensure success.

·         In an initial phase of HTE implementation the leader can establish 3rd party supplied standard screening plates for catalysis and biocatalysis.  In the mid to long term we would look to expand into customization or full internalization of plate designs.

  • The leader will provide timely updates to the SM API dev leadership team as well as the corresponding leadership teams in discovery to communicate not only successes but also advertise the capabilities available to project teams.
  • Be a member of the API DEV LT driving the department’s strategy and execution.
  • Be a member of the API Functional Scientific Review Board and the One Team forum, the two key governance bodies that drive the scientific strategy of the API DEV department
  • Represent Janssen R&D (DPDS) at external forums
  • Maintain an external network with other pharma and vendors to ensure best practices as well as remain current on developing technologies.
  • Proactively build budget requests for new capability build as well as on-boarding plans including training for staff
  • Help branding the Janssen R&D name and reputation in the API development space – academia and industry – to attract the best scientific talent

Required Knowledge, Skills and Abilities:
  • Background in High Throughput Experimentation
  • Organic synthesis, catalysis, biocatalysis
  • Knowledge of various automation capabilities (Freeslate, Quantos, FlexiWeigh, ChemScan, iChem)
  • Design of experiments, data management and visualization software
Preferred Knowledge, Skills and Abilities
  • People management
  • Experience/understanding discovery chemistry
  • Hands-on experience using various automation platforms (i.e. Mettler-Toledo FlexiWeigh and Quantos systems, iChem explorer, HEL automates, Cat96, Mosquito Liquid Dispense, TECAN, FreeSlate, EasyMax)
  • Experience/understanding in going from 96-well screening to screening reproducible at scale (ability to recognize when screening results are optimal for discovery setting versus needing for scale-up and thus more representative conditions screening)
  • Ability to explore new technologies and find value adding applications
  • Experience in tech transfer of manufacturing processes, QbD and ICH approaches to PAR and NOR.
  • AI/ML experience for data capture and mining
